The interactions between immune cells, epithelium and stressors determine the inflammation status of a host. It is widely accepted that the chronic inflammation is linked to several metabolic diseases including inflammatory bowel disease, atherosclerosis, obesity, and cancers. In the current project, we will use the normal and tumor enteroid model where enteroids mimicking the in vivo epithelial cells will be isolated from human intestine after surgical resection or from the biopsies. Several microbial products will be used as a stressor to test their effect in the integrity of the gut barrier and micronutrients will be used to determine their preventive effect. If the gut barriers are disrupted, then the harmful components can cross the epithelial layer and can interact with phagocytes including macrophages. The role of macrophages will be assessed in different disease stages. Primarily the transcriptomic and proteomic datasets will be used, and the predicted candidates will be validated in the patient-derived blood monocytes and tissue specific macrophages. This study will advance our understanding of the role of epithelial-immune cell crosstalk in chronic diseases including cancers and enhance our ability to design new targeted therapy.
